6-APB, commonly known as Benzo Fury, is an empathogenic psychoactive compound under the substituted phenethylamine and substituted benzofuran classes. It is also similar to the MDA structure with the only difference of the 3,4-methylenedioxyphenyl ring replaced with a benzofuran ring. It is also characterized as the unsaturated benzofuran derivative of the 6-APDB. 6-APB is also known to be structurally related to entactogens like MDMA, MDA, 5-APB, and 5-MAPB.

6-APB was first synthesized in 1993 as a non-neurotoxic alternative to MDMA. It did not enter the recreational market until over a decade later. The discovery of 6-APB was made by a team led by David Nichols of Purdue University. It was focused on the role of the dioxle ring structure in regulating the interaction between certain neurons. It was not known for human usage until 2010, when it was put on the market. It was very prominent in the UK’s legal highs market. In June 2013, 6-APB and other analogues were declassified as Temporary Class Drugs after the ACMD recommended that they be made Class B drugs.

Chemistry

6-APB is a 2-(2-aminopropyl)benzofuran compound. It is a member of the benzofuran class. It has an oxygen-substituted central ring and a phenyl ester group bound to an amino acid. Its core is bound to an amino group with a methyl substitution at Rα.

Initially, there were only two batches available: hydrochloride and succinate. These two batches had very different effects by weight and loose bulk densities. These were only for the treatment of certain conditions. Since then, various batches have been released, and their effects have been very different.

Pharmacology

6-APB is a reuptake inhibitor that can inhibit the reuptake of norepinephrine and other neuroactive chemicals. It can also release norepinephrine and other neuroactive chemicals. 6-APB is an alkaloid that shows higher affinity for the serotonin 5-HT 2B receptor than any other site in the world. This is because it exhibits 100-fold selectivity over the other targets.

The 6-APB protein has been found to have high affinity to the 2-adrenergic receptor subtype. This protein is not known to have clinical significance. The agonism of the 5HT2B receptor makes it more likely that 6-APB would cause long-term or chronic use in people with respiratory conditions.

With the presence of the monoamine neurotransmitters also referred to as dopamine, serotonin and noradrenaline, the brains ability to focus, plan, reward, motivate and feel pleasure is enhanced. These neurotransmitters are responsible for the brain’s ability to produce various emotions and responses. When they are inhibited or released, these molecules accumulate in the synaptic cleft and can be reused.
