Pagoclone
Pagoclone is a derivative of the cyclopyrrolone family that can be used to treat insomnia. It was produced by a team from France. It belongs to a class of non-benzodiazepines that have similar properties to those used for treating other conditions.
It binds to the binding site of human gamma-aminobutyric acid (GABA) receptors. It is a partial agonist that can be used to treat insomnia by targeting the 1, 2, and 5-containing receptors. It also has a full agonist that can be used to treat other conditions.
Compared to the parent compound, pagoclone has a greater anxiolytic activity and can induce sedation. It is also less effective at reducing the effects of sleep. In contrast, zopiclone can be used at low doses to treat insomnia.
According to David Nutt, a pharmacologist, pagoclone could be a potential base for developing a better social drug by producing positive effects on alcohol, which include sociability and relaxation. However, it can also cause negative effects, such as amnesia, nausea, and liver damage. Its effects can be reversed by the use of flumazenil, which has been used as an overdose antidote for benzodiazepines.
Studies have praised the potential of pagoclone, which was partly financed by Indevus, a company that was looking to develop a drug for treating insomnia. However, the long-term safety of the compound is not known. Based on the studies, it is believed that pagoclone could be abused similarly to diazepam, though it is less likely to cause abuse. Despite this, the development of a commercial version of the drug is still unlikely due to the concerns about abuse.
